摘要:
目的 探讨外源性H2S对糖尿病大鼠晶状体透明度的影响及其作用机制。方法 取SPF级健康雄性SD大鼠15只作为实验对象。通过腹腔注射链脲佐菌素制作糖尿病大鼠模型;按随机数字表法将大鼠随机分为3组,对照组(正常大鼠)、模型组(糖尿病模型大鼠)、治疗组(糖尿病模型大鼠+NaHS处理),每组5只。造模后,治疗组大鼠每天上午腹腔注射NaHS(5 mg·kg-1)1次,对照组和模型组给予相同体积生理盐水,连续给药12周。12周后,在田字格十字交叉处观察各组大鼠晶状体的透明度,ELISA检测各组大鼠晶状体TNF-α、AGEs的含量。结果 给药12周后,对照组大鼠晶状体透明度良好,能够清晰看到十字交叉。模型组晶状体明显混浊,看不到十字交叉。治疗组晶状体与对照组相比,出现轻微的混浊,但与模型组相比,晶状体相对透明。对照组、模型组、治疗组大鼠晶状体组织中TNF-α浓度分别为(174.91±28.44)ng·L-1、(283.75±44.46)ng·L-1、(200.14±22.15)ng·L-1,AGEs浓度分别为(302.21±49.15)ng·L-1、(470.71±56.40)ng·L-1、(369.90±67.86)ng·L-1。模型组TNF-α和AGEs浓度均高于对照组,差异均有统计学意义(均为P<0.05);治疗组TNF-α和AGEs浓度均低于模型组,差异均有统计学意义(均为P<0.05)。结论 外源性H2S能抑制TNF-α、AGEs的表达,从而提高糖尿病大鼠晶状体透明度。
Abstract:
Objective To evaluate protective effects of exogenous hydrogen sulfide (H2S) on lens opacity of rats with diabetic cataract and its underlying mechanisms.Methods Fifteen male SD rats were collected as the subjects and randomly divided into control group, model group and the treatment group (5 rats in each group). Diabetic models were reproduced by a single intraperitoneal injection of one-time chain urea with streptozotocin. After modeling, rats in the treatment group were intraperitoneally injected with NaHS (5 mg·kg-1) once every morning, and the control group and model group were given the same volume of normal saline for 12 weeks. After 12 weeks, the lens opacity of each group was observed at the cross of the Tianzi grid, and the contents of TNF-α and AGEs in the lens of each group were detected by ELISA.Results Twelve weeks after the administration, the lens opacity of the control group was well and the crisscross could be clearly seen. The lens of the model group was obviously cloudy, and the cross was not seen. Compared with the control group, the lens of the treatment group was slightly cloudy, but was relatively transparent when compared with the model group. The concentrations of TNF-α in the lens tissues of the rats in the control group, model group and treatment group were (174.91±28.44)ng·L-1, (283.75±44.46)ng·L-1, ( 200.14±22.15)ng·L-1, respectively, and the AGEs concentrations were (302.21±49.15)ng·L-1, (470.71±56.40)ng·L-1, (369.90±67.86)ng·L-1, accordingly. The concentrations of TNF-α and AGEs in the model group were higher than those in the control group, and the differences were statistically significant (both P<0.05); the concentrations of TNF-α and AGEs in the treatment group were lower than those in the model group, and the differences were statistically significant (both P<0.05).Conclusion Exogenous H2S inhibits the synthesis of TNF-ɑ and AGEs and reduce the turbidity of diabetic cataract.
