摘要:
目的 研究细胞外基质胶羊膜棒的生物学特性及其在兔干眼的应用效果。方法 使用放射免疫分析方法对细胞外基质胶羊膜棒中的表皮生长因子(epidermal growth factor,EGF)、成纤维细胞生长因子(fibroblast growth factor,FGF)、肝细胞生长因子(hepatocyte growth factor,HGF)、转化生长因子-β(transforming growth factor-β,TGF-β)等10种生物活性因子的表达含量加以测定,同时和新鲜泪道纤维蛋白胶羊膜(fresh lacrimal duct amniotic membrane,F-LDAM)、单层泪道纤维蛋白胶羊膜(monolayer lacrimal duct amniotic membrane,M-LDAM)进行对比。此外,利用负荷传感器对比细胞外基质胶泪道纤维蛋白胶羊膜(extracellular matrix lacrimal duct amniotic membrane,ECM-LDAM)与F-LDAM、M-LDAM的弹性模量、抗拉强度、断裂伸长率,并把ECM-LDAM应用于兔干眼症的治疗,术后8周观察实验兔干眼治疗前后荧光染色(fluorescein stain,FL)评分、各时段泪液分泌、FL情况和角膜共焦显微镜下角膜上皮下神经纤维密度、神经纤维分支及曲率评分。结果 ECM-LDAM、M-LDAM中EGF、NGF、HGF、TGF-β等10种因子含量均比F-LDAM低,差异均有统计学意义(均为P<0.05),而ECM-LDAM中EGF、NGF、HGF、TGF-β等10种因子含量明显高于M-LDAM,差异有统计学意义(P<0.05)。ECM-LDAM的弹性模量明显低于F-LDAM及M-LDAM,差异有统计学意义(P<0.05);而ECM-LDAM的抗拉强度和断裂伸长率明显高于与M-LDAM和F-LDAM,差异有统计学意义(P<0.05)。术后8周发现B组与A组相比FL评分降低,泪液分泌时间延长,角膜共焦显微镜下观察发现神经纤维密度、神经纤维分支及曲率评分显著降低,差异有统计学意义(P<0.05)。结论 ECM-LDAM作为一种新型安全的泪道修复材料,其丰富的生物活性因子和优越的柔韧性,可以更有效地改善兔干眼,营养修复受损的角膜神经。
Abstract:
Objective To study the biological characteristics of extracellular matrixamniotic membrane rod and its application in rabbit dry eye.Methods The expression of epidermal growth factor(EGF),fibroblast growth factor(FGF),hepatocyte growth factor(HGF),transforming growth factor-β(TGF-β)and other bioactive factors in extracellular matrix amniotic membrane rod were detected by radioimmunoassay methods and compared with fresh lacrimal duct amniotic membrane(F-LDAM) and monolayer lacrimal duct amniotic membrane(M-LDAM).Furthermore,the elastic modulus,tensile strength and elongation at break of extracellular matrix lacrimal duct amniotic membrane (ECM-LDAM) were compared between F-LDAM and M-LDAM by using load sensors.ECM-LDAM was applied to the treatment of dry eye in rabbits.Fluorescent staining (FL) score,tear secretion,FL and the nerve fiber density,nerve fiber branch and curvature score of corneal epithelium under confocal microscopy in each experimental group were observed 8 weeks before and after the operation.Results The contents of EGF,NGF,HGF,TGF-β and other factors in ECM-LDAM and M-LDAM were significantly lower than F-LDAM (P<0.05).The levels of EGF,NGF,HGF and TGF-β in ECM-LDAM and other 10 factors were significantly higher than those in M-LDAM,with statistical significance (P<0.05).The elastic modulus of ECM-LDAM was significantly lower than that of F-LDAM and M-LDAM (P<0.05),while the tensile strength and elongation at break of ECM-LDAM were higher than those of M-LDAM and F-LDAM,and the difference was statistically significant (P<0.05).At 8 weeks after operation,the fluorescence staining score of the experimental group was lower than that of the control group,and the tear secretion time was prolonged,and the nerve fiber density,nerve fiber branch and curvature score were significantly reduced in the corneal confocal microscopy.Conclusion As a new type of safe tear repair material,ECM-LDAM abundant bioactive factors and superior flexibility can improve rabbit’s dry eye more effectively as well as nourish and repair damaged corneal nerves.
