摘要:
目的 评价血管生成素(angiogenin,ANG)对小鼠急性葡萄膜炎模型眼部炎症和氧化应激的影响。方法 将BALB/c小鼠随机分为对照组、模型组和ANG组,每组各10只。ANG组小鼠给予ANG滴眼液(100 mg·L-1·d-1),对照组和模型组均给予等体积生理盐水。连续给药3 d后,模型组和ANG组在小鼠玻璃体内注射脂多糖(lipopolysaccharide,LPS)建立急性葡萄膜炎模型。125 mg·L-1 LPS给药24 h后,各组小鼠摘除眼球,收集房水,测量浸润细胞数、总蛋白浓度、一氧化氮(NO)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和氧化应激指数(oxidant stress index,OSI),并比较各组小鼠炎症反应的程度。结果 与对照组相比,模型组小鼠房水浸润细胞数、总蛋白浓度、NO、TNF-α和IL-6水平、炎症评分、OSI均显著升高(均为P<0.05),而ANG组以上各指标较模型组均显著降低(均为P<0.05)。结论 ANG可通过抑制炎症和氧化应激反应在小鼠急性葡萄膜炎模型中发挥有益作用。
Abstract:
Objective To evaluated the efficacy of angiogenin (ANG) against ocular inflammation and oxidative stress in acute uveitis model.Methods BALB/c mice were randomly divided into control group, model group and ANG group with 10 mice in each group. ANG eye drops (100 mg·L-1·d-1) were given to mice in ANG group, and equal volume normal saline was given to control group and model group. After 3 days of continuous administration, mice in the model group and ANG group were injected with lipopolysaccharide (LPS) to establish the acute uveitis model. Twenty-four hours after 125 mg·L-1 LPS administration, eyes were enucleated, and aqueous humor was collected for measurement of the number of infiltrating cells, total protein concentration, and levels of nitric oxide (NO), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and oxidant stress index (OSI). Inflammatory response severity was compared among groups clinically and histopathologically.Results Infiltrating cell number, total protein concentration, NO, TNF-α, IL-6 levels, inflammatory score, and OSI were significantly elevated in the aqueous humor of model group compared to controls (all P<0.05), while above variables in ANG group were significantly reduced compared with the model group (all P<0.05).Conclusion ANG may aid in the clinical management of uveitis by suppressing inflammation and oxidative stress.
