摘要:
目的 探讨阿魏酸对高糖诱导的人晶状体上皮细胞(human lens epithelial cells,HLEC)损伤的保护作用。方法 以HLEC为研究对象,将细胞随机分为对照组(HLEC+DMEM)、DMSO组(HLEC+DMEM+DMSO)、高糖组(HLEC+DMEM+30 mmol·L-1高糖)、高糖+阿魏酸组(HLEC+DMEM+DMSO+30 mmo·L-1高糖+阿魏酸)。采用CCK-8法检测各组细胞增殖情况;流式细胞术检测各组细胞凋亡;Q-PCR检测p53、Bcl-2、Bax mRNA表达水平;免疫细胞化学和Western blot法检测p53、Bcl-2、Bax蛋白表达水平。结果 高糖抑制HLEC的细胞活性,不同浓度阿魏酸干预后,HLEC的细胞活性均明显升高,差异均具有统计学意义(均为P<0.05)。流式细胞术检测结果显示,高糖组HLEC凋亡率高于对照组(P<0.05),高糖+阿魏酸组细胞凋亡率低于高糖组(P<0.05);高糖组p53、Bax mRNA和蛋白表达水平均明显高于对照组,而Bcl-2 mRNA和蛋白表达水平均明显低于对照组(均为P<0.05);高糖+阿魏酸组p53、Bax mRNA和蛋白表达水平均明显低于高糖组,而Bcl-2 mRNA和蛋白表达水平均明显高于高糖组(均为P<0.05)。结论 阿魏酸可能通过降低Bax和促进Bcl-2表达,进而减轻高糖诱导的HLEC损伤,从而达到防治糖尿病白内障发生的作用。
Abstract:
Objective To investigate the effects of ferulic acid on the injury of human lens epithelial cells (HLEC) induced by high glucose.Methods HLEC were randomly divided into 4 groups:Control group (HLEC+DMEM),DMSO group (HLEC+DMEM+DMSO),high glucose group (HLEC+DMEM+30 mmol·L-1 high glucose) and high glucose + ferulic acid group (HLEC+DMEM+DMSO+30 mmol·L-1 high glucose+ferulic acid).Cell proliferation was determined by CCK-8 kit,cell apoptosis by flow cytometry,mRNA expression levels of p53,Bcl-2 and Bax by Q-PCR,and protein levels of p53,Bcl-2 and Bax by immunocytochemistry and Western blot.Results High glucose solution inhibited HLEC vitality,which obviously improved by intervention of different concentrations of ferulic acid (all P<0.05).Results of flow cytometry showed that the apoptosis rate of HLEC in the high glucose group was significantly higher than those in the control group and high glucose + ferulic acid group (all P<0.05).The mRNA and proteins levels of p53 and Bax in high glucose group were significantly higher than those in control group,but the mRNA and protein levels of Bcl-2 in high glucose group was significantly lower than those in control group (all P<0.05).The mRNA and protein levels of p53 and Bax in high glucose+ferulic acid group were significantly lower than that in high glucose group,but the mRNA and protein levels of Bcl-2 in high glucose+ferulic acid group were significantly higher than those in high glucose group (all P<0.05).Conclusion Ferulic acid may protect HLEC from injury induced by high glucose through decreasing Bax and promoting Bcl-2 expression,so as for prevention and treatment of diabetic cataract.
