摘要:
目的 研究急性低压低氧环境对大鼠视网膜线粒体的影响及黄芪复方(radix astragali seu hedysari compound,RAHC) 的干预作用。方法 将42只健康SD大鼠随机分为正常氧含量对照组(常氧组)、低氧组、RAHC干预组。低氧组和RAHC干预组在低压氧舱中模拟5000 m海拔的喂养环境,RAHC组在喂养中每天予以RAHC汤药0.1 g·kg-1灌胃,低氧组予以同等剂量生理盐水灌胃。各组在上述条件处理7 d后剥离视网膜,HE染色后光学显微镜下观察大鼠视网膜组织形态变化,RT-PCR法检测锰超氧化物歧化酶(manganese superoxide dismutase,Mn-SOD)的表达,比色法测定琥珀酸脱氢酶(succinate dehydrogenase,SDH)水平,酶联免疫吸附法(ELISA)检测细胞色素C氧化酶(cytochrome c oxidase,COX)活性,免疫组化方法检测细胞色素C(cytochrome C,Cytc)的表达。结果 光学显微镜下观察发现,低氧组大鼠视网膜出现水肿,经RAHC干预后水肿减轻;低氧组Mn-SOD mRNA水平(0.811±0.057)U·mg-1较常氧组(0.950±0.920)U·mg-1下降(P<0.05),RAHC干预组Mn-SOD mRNA水平(0.884±0.101)U·mg-1较低氧组高,差异有统计学意义(P<0.05);低氧组SDH水平(22.009±0.384)U·mgprot-1较常氧组(30.104±0.854)U·mgprot-1低(P<0.05),且RAHC干预后无明显差异(P>0.05);低氧组COX水平(0.632±0.031)ng·mL-1较常氧组(2.461±0.097)ng·mL-1低(P<0.01),RAHC干预组COX水平(1.204±0.037)ng·mL-1较低氧组高,差异有统计学意义(P<0.01)。与常氧组相比,低氧组大鼠Cytc在视网膜神经节细胞层、神经纤维层、内丛状层及外丛状层中棕色染色阳性表达增强,但在RAHC干预组中的表达强度减弱。结论 急性低压低氧环境导致视网膜线粒体发生损伤,RAHC对急性低压低氧导致的视网膜损伤有保护作用,可能与上调Mn-SOD、COX活性及减少Cytc的释放有关。
Abstract:
Objective To explore the effect of acute hypobaric hypoxia on retinal mitochondria and intervention effect of Radix Astragali seu Hedysari Compound (RAHC) on retinal mitochondrion injury in rats.Methods Totally 42 healthy SD rats were randomly divided into 3 groups:Normoxic control group (normoxia group),hypoxic group and RAHC intervention group.Rats in hypoxic group and RAHC intervention group were fed at a hypobaric oxygen chamber (simulate 5000 m altitude).Rats in RAHC group was given 0.1 g·kg-1 RAHC compound decoction by gavage each day,and those in hypoxic group received same amount of normal saline by gavage.The retinas were collected after different treatments for 7 days.After HE staining,morphological changes of retinal tissues were observed under optical microscope.The expression of manganese superoxide dismutase (Mn-SOD) was detected by RT-PCR,level of succinate dehydrogenase (SDH) was determined by colorimetry,activity of cytochrome c oxidase (COX) was measured with enzyme-linked immunosorbent assay (ELISA),and expression level of cytochrome C (Cytc) was detected by immunohistochemistry.Results Edema was found in the retina of rats in hypoxia group under light microscope,and was alleviated after the intervention of RAHC.The Mn-SOD mRNA levels were (0.811±0.057)U·mg-1 in the hypoxia group,and lower than (0.950±0.920)U·mg-1 in normoxic control group and (0.884±0.101)U·mg-1 in RAHC intervention group,statistical differences were observed (all P<0.05).SDH levels were (22.009±0.384)U·mgprot-1 in the hypoxic group,and lower than (30.104±0.854)U·mgprot-1 in normoxic control group (P<0.05).There was no obvious change of SDH level after RAHC intervention (P>0.05).COX levels were (0.632±0.031)ng·mL-1 in hypoxic group,lower than (2.461±0.097)ng·mL-1 in normoxic control group and (1.204±0.037)ng·mL-1 in RAHC intervention group (all P<0.01).Compared with normoxic control group,positive brown staining of Cytc increased in retinal ganglion cell layer,retinal nerve fiber layer,inner plexiform layer and outer plexiform layer of rats in hypoxic group,and decreased in RAHC intervention group.Conclusion Acute hypobaric hypoxia can cause retinal mitochondrion injury,while RAHC exerts protective effects on the injury by upregulating Mn-SOD and COX activities as well as reducing the Cytc release.
