摘要:
多种疾病的发生发展均与慢性低度炎症反应有关,其中免疫异常起重要作用。核苷酸结合寡聚化结构域样受体蛋白3(nucleotide-binding oligomerization domain-like receptor protein 3,NLRP3)炎症体作为固有免疫系统的重要组成成分,已被证实在糖尿病、冠状动脉粥样硬化性心脏病、风湿免疫疾病及肿瘤等多种疾病中发挥重要作用。NLRP3炎症体可被多种代谢信号激活,进而引起caspase-1的活化,最终导致细胞释放IL-1β和IL-18,诱发炎症级联反应,引发组织损伤。近来多项研究表明,NLRP3炎症体可能成为多种疾病新的治疗靶点。本文将对NLRP3炎症体的活化与调控机制及其在年龄相关性黄斑变性、角膜炎、葡萄膜炎、青光眼、干眼症、糖尿病视网膜病变及缺血再灌注损伤等眼科疾病中的研究进展及相关治疗前景进行综述。
Abstract:
Chronic low-grade inflammation has been proven to be involved in the occurrence and development of many diseases,and the immune abnormity plays an important role.As an important part of innate immune system,nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome plays a prominent role in diabetes,coronary diseases,rheumatic immune diseases and tumors.Once stimulated by many metabolicsignals,NLRP3 inflammasome can activate caspase-1 and further lead to the release of interleukin-1 β(IL-1β) and interleukin-18(IL-18).This may initiate a cascade of inflammation and cause tissue damage.Plenty of recent researches have conclusively proven NLRP3 inflammasome to be a new therapy target for many diseases.In this paper,we will make a review for the activating pathway and mechanism of NLRP3 and its role in age-related macular degeneration,keratitis,uveitis,glaucoma,xerophthalmia,diabetic retinopathy and retinal ischemia/reperfusion injury ect.The related therapeutic potentials are also summarized.
