摘要:
目的 观察角膜基质注射脐带间充质干细胞(umbilicalcordmesenchymalstemcells,UMSCs)治疗兔角膜碱烧伤的效果。方法 健康新西兰大白兔随机分为3组,每组12只,每只兔左眼为角膜碱烧伤模型眼。A组兔左眼在碱烧伤2d后角膜基质内注射含有2×106个UMSCs的磷酸盐缓冲液2μL,B组注射相同剂量的磷酸盐缓冲液,C组为角膜碱烧伤后未进行处理组,在注射后不同时间对角膜混浊程度、新生血管生长情况、角膜上皮缺损情况等进行临床观察并评分,同时进行综合评分。结果 注射后14dA组新生血管生长较B组明显缓慢,A组、B组注射隧道周围角膜新生血管生长评分分别为0分、2分,差异有统计学意义(P<0.05)。注射后2d和6dA组角膜混浊程度较B组明显轻,注射后2dA组、B组角膜混浊程度评分分别为2分、4分,注射后6d评分与2d相同,差异均具有统计学意义(均为P<0.05)。注射后2d、7d角膜上皮荧光素染色评分各组之间差异均无统学意义(均为P>0.05)。结论 碱烧伤后角膜基质注射UMSCs可减少新生血管的形成,为抑制碱烧伤后角膜血管化提供参考,碱烧伤后角膜基质注射UMSCs可在一定程度上恢复角膜透明度。
Abstract:
Objective To evaluate the efficacy of stromal injection of umbilical cord mesenchymal stem cells ( UMSCs) after rabbit corneal alkali burn. Methods Corneal alkali injury models were established in left eyes of healthy New Zealand rabbits .which were divided into three groups , 12 rats in each group. Group A was treated with intrastromal injections of phosphate-buffered saline ( PBS) 2 VL containing 2 x 106 UMSCs. Group B was treated with PBS by the same applications forms. Group C did not give any treatment after corneal alkali burn. Rabbits were evaluated for corneal neovascularization , opacification and epithelial defects at 2 days , 3 days ,6 days ,7 days , 14 days after injection. and scored on it according to the criteria, a comprehensive score was taken at the same time. Results At 14 days after injection , the growth of corneal neovascularization in group A was much slower than group B ,the mean corneal neovascularization area score in group A and group B was o points and 2 points ,respectively , the difference was statistically significant (P < 0. 05 ) . At 2 days and 6 days after injection, the corneal opacity of group A was less severe than group B , the corneal opacity score in group A and group B was all 2 points and 4 points,the differences were statistically significant ( all P < 0. 05 ) . Corneal fluorescence staining scores at 2 days ,7 days after injection between each group had no statistical significance ( P > 0. 05 ) . Conclusion Intrastromal injection of UMSCs after alkali burn cornea can reduce the formation of angiogenesis , which can provide a reference for the inhibition of corneal revascularization after alkali burn , and restore corneal transparency in a certain extent.
