摘要:
 目的 探索京尼平诱导兔眼巩膜胶原交联的有效性和安全性,为防治近视进展寻找有效的方法。方法 20只成年新西兰大白兔,按简单数字随机方法选一眼Tenon’s囊下注射0.5mmol?L-1京尼平0.5mL2次和4次各10只,再按简单数字随机选对侧眼10只为对照眼;每组5只测试生物力学,另5只进行组织学检查。结果 与对照眼比较,Tenon’s囊下注射2次和4次,巩膜的极限应力分别增加了95%和161%;同时20%应变时弹性模量分别增加了204%和260%,而极限应变则分别下降了13%和22%。京尼平没有引起巩膜、脉络膜和视网膜中的细胞凋亡,也没有损伤邻近组织角膜和视神经,治疗区域没有明显的颜色变化。结论 京尼平诱导兔眼巩膜胶原交联安全有效,是一种潜在的防治近视进展的方法。
Abstract:
 Objective To evaluate the efficiency and safety of crosslinking scleral collagen using genipin in rabbit . and develop new therapy for preventing the progression of myopia. Methods Twenty New Zeal rabbits were randomly divided into two groups (n = 10) for sub-Tenon ’ s injection of 0. 5 mmol . L-l genipin for 2 times and 4 times . and the contralateral eyes were randomly selected 10 eyes as control. Five eyes in each group were for stress-strain measurements .and other 5 eyes were prepared for histological examination. Results Compared with untreated sclera strips , after sub-Tenon injecting with genipin for 2 times and 4 times. the ultimate stress of genipin crosslinked sclera increased by an amplitude of 95% and 161% . The Young ’ s modulus at 20% strain increased by 204% and 260% , respectively. And ultimate strain decreased by 13% and 22% . It did not cause apoptosis in scleral , choroid and retinal cells ; and no side effects were found in the peripheral cornea and optic nerve. There had no visible discoloration around treated area. Conclusion Genipin-induced collagen crosslinking is effective and safe in rabbit , and is a potential agent to strengthen the weaken sclera to prevent progressive myopia.
