摘要:
 目的 探讨卡铂联合MicroRNA-34a对视网膜母细胞瘤HXO-RB44细胞的增殖抑制作用和对裸鼠异位瘤的生长抑制作用。方法 将HXO-RB44细胞分成卡铂干预组、MicroRNA-34a干预组、联合干预组和生理盐水对照组。MTT检测卡铂联合MicroRNA-34a干预对HXO-RB44细胞的毒性。流式细胞技术检测卡铂联合MicroRNA-34a干预诱导HXO-RB44细胞凋亡和对细胞周期的影响。构建裸鼠视网膜母细胞瘤异位肿瘤模型,检测卡铂联合MicroRNA-34a干预对裸鼠肿瘤的生长抑制能力。结果 给药48h后,HXO-RB44细胞的存活率:分别为卡铂干预组(58.5±4.4)%、MicroRNA-34a干预组(64.3±3.1)%、联合干预组(27.8±2.2)%,差异有统计学意义(P<0.01);HXO-RB44细胞的凋亡率:分别为卡铂干预组(13.5±1.4)%、MicroRNA-34a干预组(8.2±1.1)%、联合干预组(31.5±2.2)%,差异有统计学意义(P<0.01);联合干预组较生理盐水对照组S期细胞增加,G0/G1期细胞减少,G2/M期细胞减少(均为P<0.01);荷瘤裸鼠治疗实验结果显示卡铂干预组、MicroRNA-34a干预组和联合干预组的肿瘤生长抑制率分别为44.6% ±1.3%、36.5% ±1.3%和75.3% ±1.5%,与生理盐水对照组差异均有统计学意义(均为P<0.01)。结论 卡铂联合MicroRNA-34a能够有效抑制HXO-RB44细胞的增殖和肿瘤的生长。
Abstract:
 Objective To evaluate the effects of Carboplatin combined with MicroRNA-34a on the inhibition of HXO-RB44 cells proliferation and tumor growth. Methods The HXO-RB44 cells were divided into Carboplatin group , MicroRNA-34a group , Carboplatin + MicroRNA-34a group and saline group. The MTT assay was used to evaluate the anti-proliferation efficiency of Carboplatin combined with MicroRNA-34a. Flow cytometry was used to evaluate the HXO-RB44 cells induced apoptosis in Carboplatin + MicroRNA-34a group. HXO-RB44 cells were xenografted in mice to establish the animal model, which were used to evaluate the effect of anti-tumor. Results The cell viability of HXO-RB44 cells in Carboplatin group , MicroRNA-34a group and Carboplatin + MicroRNA-34a group were ( 58. 5 + 5. 2 ) % , ( 64. 3 + 7. I ) % . ( 27. 8 + 3. 2 ) % , respectively , there was significant difference (P < 0. 01 ). The apoptotic rate of HXO-RB44 cells in Carboplatin group, MicroRNA-34a group and Carboplatin + MicroRNA-34a group were ( 13. 5 + 1. 4 ) % , ( 8. 2 + 1. 1) % . ( 31. 5 +2. 2) % , respectively , there was sigruficant difference (P < 0. 01) . Compared with the saline group , the cells in S phase in Carboplatin + MicroRNA-34a group increased , the cells in GO/GI phase decreased, and the cells in G2/ M phase also decreased ( all P < 0. 01 ) . The inhibitive rate of tumor growth in Carboplatin group , MicroRNA-34a group and Carboplatin + MicroRNA-34a group were ( 44. 6 + 1. 3 ) % . ( 36. 5 + 1. 3 ) % , ( 75. 3 + 1. 5 ) % , respectively , there was significant difference compared with the saline group ( all P < 0. 01 ) . Conclusion Carboplatin combined with MicroRNA-34a can effectively inhibit the HXO-RB44 cells proliferation and tumor growth.
