摘要:
 目的 观察人脐血干细胞(humanumbilicalcordbloodstemcells,hUCBSC)移植对视神经部分受损SD大鼠视网膜神经节细胞(retinalganglioncells,RGC)的保护作用。方法 将48只健康成年SD大鼠随机分为2组,2组SD大鼠暴露视神经,应用40g夹持力的视神经夹在大鼠眼球后2mm处夹视神经30s造成部分视神经损伤模型,均损伤左眼。A组:伤后1周玻璃体腔注射脐血干细胞载体PBS,24只。B组:伤后1周玻璃体腔注射hUCBSC,24只;2组均于注射后7d、14d、21d、28d处死动物。处死前7d双上丘注射50g?L-1荧光金逆行标记双眼RGC。然后按时间先后处死大鼠分离视网膜置于荧光显微镜下,计数2组RGC并计算RGC标识率。结果 A、B两组视神经损伤眼RGC计数均低于未损伤眼(均为P<0.05),且随时间延长两组RGC标识率均呈下降趋势(均为P<0.05),但B组注射后7d、14d、21d、28dRGC标识率(77.52±6.33)%、(74.12±8.23)%、(64.78±5.21)%、(59.93±9.00)%下降幅度明显较A组(75.68±8.74)%、(68.21±10.59)%、(56.24±9.34)%、(48.91±8.81)%平缓。同时间段B组RGC标识率均高于A组,且差异均有统计学意义(均为P<0.05)。结论 玻璃体腔注射hUCBSC可减缓外伤性视神经损伤大鼠视网膜中RGC的凋亡,对RGC具有一定的保护作用。
Abstract:
 Objective To observe the protective effects of human umbilical cord blood stem cells ( hUCBSC) vitreous transplantation on retinal ganglion cells ( RGC) in the Spragur-Dawly rats with optic nerve injury. Methods Forty-eight adult Sprague-Dawley rats were randomly divided into two groups, 24 rats in each group. Calibrated optic nerve crush injury model was induced in the left eyes in the six groups , the right eyes served as control. Medicine was injected at 7 days after optic nerve injury. PBS was injected into vitreous of group A. The hUCBSC were injected into vitreous of group B. And then the rats were sacrificed at 7 days,14 days ,21 days and 28 days. Seven days before sacrifice ,50 g . L -l fluorogold was injected into superior colliculi bilaterally to label the RGC. The eyes were enucleated. and flat mounts of the retina from both eyes were prepared on a slide and observed under a fluorescence microscope. The labeled RGC were counted by a computerized image analyzer, and the labeled RGC rate was used for statistical analysis. Results The number of RGC in the injury eye was less than that in the control eye in group A , B ( all P < 0. 05 ) . The labeled RGC rate showed a declined tendency gradually along with increases of the time in two groups ( all P < 0. 05 ) , but the decreased trend of group B, which were ( 77. 52 + 6. 33 ) % at 7 days, ( 74. 12 + 8. 23 ) % at 14 days , ( 64. 78 +5. 21) % at 21 days and (59. 93 +9. 00) % at 28 days ,were evidently slower than those of group A, which were ( 75. 68 + 8. 74) % at 7 days , ( 68. 21 + 10. 59) % at 21 days , ( 56. 24 + 9. 34) % at 24 days and (48. 91 +8. 81) % at 28 days , there were statistical differences between two groups ( all P < 0. 05 ) . Conclusion The vitreous injection of hUCBSC can decrease the apoptosis of RGC in ratina of rats with traumatic optic nerve injury , and has the protective effect on RGC.
