摘要:
目的 研究线粒体融合蛋白基因2(mitofusin 2,Mfn2)在转化生长因子β2(transforming growth factor β2,TGF-β2)诱导的人晶状体上皮细胞(human lens epithelial cells,HLECs)发生上皮-间质转化(epithelial-mesenchymal transition,EMT)过程中的表达及作用。方法 实时荧光定量PCR检测不同浓度(10 ng·mL-1、50 ng·mL-1、100 ng·mL-1)TGF-β2处理HLECs 24 h后,各TGF-β2处理组与对照组(0 ng·mL-1 TGF-β2) Mfn2 mRNA表达量差异。选取最佳作用浓度后,免疫荧光细胞化学检测TGF-β2处理组与对照组的Mfn2蛋白表达变化,实时荧光定量PCR和免疫荧光细胞化学检测分别验证TGF-β2处理组与对照组中E-钙黏蛋白(E-Cadherin)和波形蛋白(Vimentin) mRNA表达量和蛋白表达变化。Mfn2基因荧光表达载体(pEGFP-Mfn2)转染后实时荧光定量PCR和免疫荧光细胞化学检测分别验证转染组和对照组Mfn2、E-Cadherin和Vimentin mRNA表达量差异和蛋白表达变化。结果 TGF-β2处理组与对照组相比,E-Cadherin mRNA表达量减少;Vimentin、Mfn2的mRNA表达量均增加,且差异均有统计学意义(均为P<0.001)。E-Cadherin蛋白表达降低,Vimentin、Mfn2的蛋白表达增加。成功转染Mfn2后转染组与对照组相比,E-Cadherin mRNA表达量下调,Vimentin mRNA表达量均增加,差异均有统计学意义(均为P<0.001)。E-Cadherin蛋白表达降低,Vimentin蛋白表达增加。结论 HLECs在发生EMT时Mfn2表达量上调,过表达Mfn2可使HLECs发生EMT。Mfn2参与了TGF-β2诱导的HLECs的EMT过程。
Abstract:
Objective To investigate the expression and role of mitofusin 2 (Mfn2) in the epithelial-mesenchymal transition (EMT) of human lens epithelial cells (HLECs) induced by transforming growth factor β2 (TGF-β2).Methods Real-time fluorescence quantitative PCR is used to detect the difference in mRNA expression levels of Mfn2 between the TGF-β2 treated group and the control group (0 ng·mL-1) after HLECs was treated with TGF-β2 for 24 h at different concentrations (10 ng·mL-1, 50 ng·mL-1, 100 ng·mL-1). After the optimal concentration was selected, the Mfn2 protein expression in the TGF-β2 treated group and control group was detected by immunofluorescence cytochemistry. Real-time fluorescence quantitative PCR and immunofluorescence cytochemical detection were used to verify the changes in mRNA expression and protein expression of E-Cadherin and Vimentin in the TGF-β2 treated group and control group, respectively. After transfection of the Mfn2 gene fluorescent expression vector (pEGFP-Mfn2), the expression changes of mRNA and protein for Mfn2, E-Cadherin and Vimentin in the transfection group and the control group were verified by real-time quantitative PCR and immunofluorescence cytochemical detection, respectively.Results Compared with the control group, the expression of E-Cadherin mRNA decreased in the TGF-β2 treated group; the expression levels of Vimentin and Mfn2 mRNA increased, and the differences were statistically significant (both P<0.001). The expression of E-cadherin decreased, and the expression of Vimentin and Mfn2 protein increased.Compared with the control group, the expression of E-Cadherin mRNA decreased and expression of Vimentin mRNA increased in transfection group after successful transfection of Mfn2, and statistical differences were found (both P<0.001). Expression of E-Cadherin protein decreased and Vimentin protein increased.Conclusion The expression of Mfn2 is up-regulated when EMT occurs in HLECs. Overexpression of Mfn2 can induce EMT in HLECs. Mfn2 is involved in the EMT process of TGF-β2-induced HLECs.
