摘要:
目的 探讨组织因子靶向肽(tissue factor targeting peptide,TF-TP)对激光诱导C57BL/6小鼠脉络膜新生血管(choroidal neovascularization,CNV)形成的影响。方法 选取36只清洁级C57BL/6雌性小鼠分为3组:正常对照组、激光组和TF-TP给药组。激光光凝后第1天开始,TF-TP给药组每隔1 d应用5 μL微量进样器给予小鼠玻璃体内注射 1 μL 浓度为100 mg·L-1的TF-TP,共3次。通过FFA评价各组小鼠CNV渗漏情况;采用组织病理学方法观察光凝后14 d各组小鼠CNV中央最大厚度;采用免疫组织化学法测定视网膜脉络膜组织中CD34的表达;通过Western blot检测小鼠RPE-脉络膜复合物中TF蛋白的表达。结果 激光组小鼠在光凝后14 d,视网膜光斑处可见明显荧光渗漏点,造影晚期呈弥漫性荧光扩散,总渗漏率为69.79%。TF-TP给药组小鼠视网膜光斑处可见轻度荧光渗漏,造影晚期可见渗漏处高荧光点但无扩散,总渗漏率为55.21%。TF-TP给药组的渗漏率低于激光组,差异有统计学意义(P=0.037)。TF-TP给药组CNV中央最大厚度为(60.02±2.48)μm,明显低于激光组(115.89±5.04)μm,差异有统计学意义(P<0.05)。TF-TP给药组视网膜脉络膜组织中CD34表达的A值为0.09±0.03,明显低于激光组(0.16±0.01),差异有统计学意义(P<0.05)。正常对照组、激光组、TF-TP给药组TF蛋白的相对表达量分别为0.14±0.01、0.40±0.02、0.22±0.03。TF-TP给药组TF蛋白的相对表达量明显低于激光组,差异有统计学意义(P<0.05)。结论 TF-TP可以减小激光诱导的小鼠CNV厚度,降低视网膜脉络膜组织中CD34以及RPE脉络膜混合物中TF蛋白的表达,抑制小鼠CNV的形成。
Abstract:
Objective To investigate the effect of tissue factor targeting peptide (TF-TP) on C57BL/6 mice choroidal neovascularization(CNV) induced by laser.Methods Thirty-six SPF C57BL/6 female mice were divided into 3 groups:normal control group,laser group and TF-TP group.From the first day after laser injury,the intravitreal injection of 1 μL of TF-TP (100 mg·L-1) was carried out in mice of the TF-TP group every other day for a total of 3 times.Fundus fluorescein angiography (FFA) was used to measure the CNV leakage degree.The maximum thickness of CNV was observed by histopathological examination on day 14 after laser photocoagulation.The expression of CD34 was detected by immunohistochemical staining.The expression of TF in RPE-choroidal complex of mice was measured by Western blot.Results On day 14 after photocoagulation,FFA showed that there was obvious fluorescence leakage at the retina of the laser group,and the diffuse fluorescence was diffused at the later stage,and the leakage rate was 69.79%.In the TF-TP group,mild fluorescence leakage was observed,and the high fluorescence but no diffusion was detected at the late stage,and the leakage rate was 55.21%.Compare with the laser group,the leakage rate of the TF-TP group was significantly decrease(P=0.037).The CNV thickness in TF-TP group[(60.02±2.48)μm] was significantly lower than that in the laser group[(115.89±5.04)μm](P<0.05).The average optical density of CD34 in the TF-TP group[(0.09±0.03)]was significantly lower than that of the laser group(0.16±0.01),with a significant difference between two groups(P<0.05).The relative expression of TF protein in the normal control group,the laser group and the TF-TP group were 0.14±0.01,0.40±0.02 and 0.22±0.03,respectively.Compared with the laser group,the expression of TF protein were significantly decreased in the TF-TP group(P<0.05).Conclusion TF-TP can reduce the CNV thickness in the laser-induced mouse,and down-regulate the expression of CD34 and the level of TF protein,which may inhibit the formation of CNV in mice.
