摘要:
 目的 研究那他霉素粉末直接给药和滴眼液给药在兔眼角膜的药代动力学的差异。方法 将16只新西兰白兔随机分为A、B两组,每组8只,每只白兔左右眼分别使用那他霉素干粉(每10min涂1次)和那他霉素眼液(每5min滴20μL)。2h后A组取其角膜,B组抽取房水。运用高效液相色谱法分别测定那他霉素含量,柱温为30℃,流动相为乙酸铵溶液-乙腈-四氢呋喃,流速为1mL?min-1,检测波长为303nm。结果 在该色谱条件下,那他霉素的保留时间为6.5min,分离度良好,标准曲线在0.05~50.00mg?L-1内线性关系良好(r=1.000),最低定量限为0.05mg?L-1。A组经那他霉素粉末给药的角膜中药物浓度为(19.39±1.95)mg?L-1,较滴眼液给药组的(10.34±3.02)mg?L-1显著增高,差异有显著统计学意义(P<0.001);B组经粉末给药的房水中药物浓度为(1.67±0.15)mg?L-1,较滴眼液给药组的(1.46±0.14)mg?L-1明显增高,差异有统计学意义(P=0.013)。结论那他霉素粉末在兔眼角膜的穿透性高于那他霉素滴眼液。
Abstract:
 Objective To study the differences of pharmacokinetics in the rabbit cornea of natamycin powder administration directly and eye drops administration. Methods Sixteen New Zealand rabbits were randomly divided into two groups , each group consisted of 8 rabbits. The left and right eye of every rabbit was adnunistrated with natamycin eye drops (20 VL per 5 nunutes) and natamycin powder ( once per 10 minutes) respectively. The corneal tissue in group A was taken , and the aqueous humor in group B was extracted. Natamycin content was determined by high performance liquid chromatography(HPLC). The column temperature was 30 9C . the mobile phase of ammonium acetate solution-acetonitrile-tetrahydrofuran, the flow rate was I mL . min’1 ,the detection wavelength was 303 nm. Results Under the chromatographic conditions,natamycin retention time was 6. 5 minutes, separation was good, standard curve within 0. 05 - 50. 00 mg . L-I had good linear relationship ( r = 1. 000 ) , limit of quantification was 0. 05 mg . L -’. Natamycin concentration in cornea adnunistrated with powder [ ( 19. 39 + 1. 95 ) mg . L-l ] was much higher than that administrated with eye drops [ ( 10. 34 + 3. 02) mg . L -l ] (P < 0. 001 ) ; Natamycin concentration in aqueous humor adnunistrated with powder [ ( 1. 67 +0. 15 ) mg . L-l ] was also higher than that administrated with eye drops [ (1. 46 +0. 14) mg . L-I ] (P = 0. 013). Conclusion The penetrability of natamycin powder in rabbit cornea is higher than that of eye drops.
