摘要:
 目的 观察细胞型朊蛋白(cellularprionprotein,PrPc)在2周龄、9个月龄、18个月龄正常ICR小鼠视网膜中的表达情况。方法 选取2周龄、9个月龄、18个月龄ICR小鼠各20只,取出视网膜分别使用RT-PCR检测视网膜组织中PrPc基因的表达情况,Westernblot检测小鼠视网膜组织中PrPc蛋白的表达情况。分析小鼠视网膜中PrPc的表达水平与月龄的关系及18个月龄小鼠视网膜与脑组织中PrPc的表达差异。结果 2周龄、9个月龄、18个月龄小鼠视网膜组织中PrPc基因表达的相对光密度值分别为0.175±0.010、0.420±0.044、0.692±0.030,两两比较,差异均有统计学意义(均为P<0.05);18个月龄小鼠脑组织中PrPc基因表达的相对光密度值为1.363±0.255,与18个月龄小鼠视网膜组织中PrPc基因表达的相对光密度值比较,差异有统计学意义(P<0.05)。2周龄、9个月龄、18个月龄小鼠视网膜组织中PrPc蛋白表达的相对光密度值分别为0.319±0.028、0.698±0.066、0.996±0.058,两两比较,差异均有统计学意义(均为P<0.05);18个月龄小鼠脑组织中PrPc蛋白表达的相对光密度值为2.008±0.192,与18个月龄小鼠视网膜组织中PrPc蛋白表达的相对光密度值比较,差异有统计学意义(P<0.05)。结论 正常小鼠视网膜中存在PrPc的表达,且表达水平随着小鼠月龄的增加而逐渐增强,但弱于脑组织中的PrPc表达强度。
Abstract:
 Objective To observe the expression of cellular prion protein ( PrP’ ) in retina of ICR mice with 2 weeks old.9 months old and 18 months old. Methods The ICR mice with 2 weeks old.9 months old and 18 months old were chosen,20 cases in each month age ,the retina were extracted.and the expression of Prpe gene and protein were detected by RT-PCR and Western blot. The relationship between expression levels of PrP’ in the mouse retina and month age was analyzed, and the difference of Prpe expression rn retina and brain tissue of ICR mice with 18 months old was also analyzed. Results The relative optical density value of Prpe gene expression in retina of mice with 2 weeks old,9 months old and 18 months old were 0. 175 + 0. 010 .0. 420 + 0. 044 and 0. 692 + 0. 030 , respectively, there were statistical differences between each two month age ( all P < 0. 05 ) . The relative optical density value of Prpe gene expression in brain tissue of ICR mice with 18 months old was l. 363 +0. 255 , there was statistical difference compared with Prpe gene expression in retina (P < 0. 05 ) . The relative optical density value of Prpe protein expression in retina of mice with 2 weeks old.9 months old and 18 months old were 0. 319 +0. 028 ,0. 698 +0. 066 and 0. 996 + 0. 058 , respectively , there were statistical differences between each two month age ( all P < 0. 05 ) . The relative optical density value of PrPe protein expression in brain tissue of ICR mice with 18 months old was 2. 008 + 0. 192 . there was statistical difference compared with Prpe protein expression in retina (P < 0. 05 ) . Conclusion The expression of PrPe exists in normal mouse retina.and the expression level increase with increasing month age of mice , but the expression in retina is weaker than that in brain tissue.
